In practice, it is a cost and feasibility decision that should be made before a single prototype is produced. Each route changes three things at once: the overage you must add to hit the label claim at end of shelf life, the true cost per dose of viable cells, and whether the formula can physically be made into a chewable tablet, a gummy or a stick sachet. Get the order wrong—choose the format first, then ask the powder to survive it—and the rework lands on stability studies, artwork and tooling.

Probiotic viability is lost at three separate gates, and no protection technology covers all of them. The first gate is shelf life: moisture and oxygen ingress degrade cells in the finished pack. The second is gastric transit: acid and bile reduce the number of cells that arrive intact. The third is manufacturing: compression force, depositing heat, shear, extrusion and heat sealing all take a toll before the product is even packed. Free, unprotected powder offers no barrier at any gate; it relies entirely on the strain and on tight control of water activity and packaging. Microencapsulation—alginate beads, fat matrices, protein or polysaccharide coatings—mainly addresses the first and third gates by separating cells from moisture, oxygen and mechanical stress. Enteric coating, whether applied to the capsule shell or to granules, mainly addresses the second.

Before buying protection, it is worth separating what the strain already does from what the coating adds. Some strains survive acid and processing well on their own; paying for encapsulation on top of a robust strain can be money spent twice. Conversely, no coating reliably rescues a strain that is intrinsically fragile or was chosen for a claim it cannot support at the required dose. The two technologies also interact: an enteric-coated capsule protects the powder inside it from acid but does nothing for a gummy's moisture, while a well-encapsulated powder still dissolves in the stomach unless it sits inside an enteric shell. They can be stacked—encapsulated powder in an enteric capsule—but only if the capsule's disintegration behaviour and the coating's moisture barrier are tested together, not assumed.

Supplier language tends to outrun the evidence. 'Acid resistant', 'heat stable' and 'protected by double coating' are claims, not specifications. The verifiable version names the assay: which strain, in what matrix, at what pH and bile concentration, sampled at which time points, plated on which medium, with what recovery method. A survival percentage quoted without those parameters cannot be compared with a competitor's number, and should not be used to justify a lower overage.

When you request data, ask for simulated digestion results that state the strain, the carrier or food matrix, the time points and the enumeration method—otherwise two 'high survival' figures may describe completely different experiments. Then ask for processing stress data specific to your intended format. For chewables and swallowable tablets, that means viability measured before and after compression at the forces your tooling will actually run. For gummies, depositing temperature and the water activity of the finished piece matter more than any generic heat claim. For stick sachets, the losses come from extrusion, dosing and the heat-seal zone; ask what the powder's particle size distribution does to flow, dusting and dispersion in the stick.

Stability data should arrive as a curve, not a single end-point. Ask for the storage conditions (temperature and relative humidity), the sampling intervals, and the measured viable count at each point, for both accelerated and real-time studies, generated on the finished format rather than the raw powder. On the specification and COA, expect the items that make the technology auditable: encapsulation rate and the method behind it, water activity, particle size distribution, strain identity confirmation, and a reference for each method used. If a third-party report is offered, check that the sample description, batch number and test dates line up with the material you would actually receive.

Price per kilogram of powder is not a comparable number. Convert every quote to cost per dose of viable cells at end of shelf life: purchase price divided by the viable count you can actually claim, adjusted for the overage required, process losses and rejected batches. This is where encapsulation often pays for itself—if it lets you cut overage substantially and raises the probability of passing shelf-life testing on the first attempt, a higher unit price can still produce a lower cost per dose. Run the same arithmetic for the unprotected option and see whether the saving survives the extra overage and the risk of a failed stability run.

The format also constrains the technology. High-compression tableting punishes unprotected cells and even some brittle coatings; low-moisture gummy systems demand a carrier that does not migrate water; stick sachets reward free-flowing, low-dust powders and tolerate coatings only if they disperse cleanly in the glass. Beyond unit cost, count MOQ, the number of qualified suppliers for the chosen powder, and switching cost: changing the powder source or coating process usually means re-running stability and re-validating the label claim.

Sequence the decision deliberately: market and shelf-life target first, then format, then strain and dose, then the protection technology—and only then the supplier quote.

At sampling, run three validation sets in parallel rather than in sequence: processing tolerance on the real equipment, accelerated stability on the packed format, and sensory and appearance checks after storage. Agree in advance what counts as a pass for each, so a marginal result is not negotiated after the fact. In the supply agreement, make powder source, coating process and manufacturing site change-controlled, require re-validation and retained samples, and state who owns and pays for the stability re-check. A short list of questions—what method was used, on which strain, in which matrix, backed by which batch record—will separate suppliers with real data from those with a brochure.
","Microencapsulation vs Enteric Coating: Probiotic Delivery","Compare free powder, microencapsulation and enteric coating by overage, cost per viable dose and format
